Abstract

Begonia x hiemalis Fotsch. (Elatior Begonia) is known as a temperate ornamental plant, which is commercially used as a flowering pot plant. To better understand in vitro plant regeneration, the process of somatic embryogenesis was examined and scanning electron microscopy was performed on Begonia. Direct somatic embryogenesis induction of Begonia x hiemalis Fotsch. var. Scwabenland Red was done using leaf explants. The explants were treated with 1.0 mg/L TIBA and cultured on MS medium supplemented with 1.0 mg/L benzylaminopurine (BAP), 0.1 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D), and 500 mg/L casein hydrolysate. Cultures were incubated in the dark for 24 hours. Different stages of somatic embryos were observed in the cultures. The globular stage was observed after six to seven weeks incubation and the heart-shaped, torpedo and cotyledonary stages emerged after subculturing the explants on MS medium supplemented with 0.5 mg/L gibberelic acid (GA3). Morphological studies of the different stages of somatic embryogenesis of Begonia were observed under scanning electron microscope (SEM). The cotyledonary stage was then transferred to regeneration medium for embryogenic maturation and regeneration.
